RESUMEN

The present article aims to evaluate the antifungal and antivirulence effect of the phytoconstituent Limonene against Candida spp. Antifungal assays were performed, where the concentration capable of inhibiting 50 % of fungal growth, the growth inhibition curve, the minimum fungicidal concentration, the evaluation of the modifying effect with fluconazole, the inhibitory effect of the substances on the morphological transition of Candida spp. and the statistical analysis of the results were determined. With this study, it was seen that limonene demonstrated growth inhibition for the strains tested and when associated the natural compound with Fluconazole, there was potentiation of the effect of the drug, since the inhibition of growth by the combination occurred at lower concentrations against all strains tested, when compared to the drug alone, which inhibited growth at the highest concentration. In the test to determine the Minimum Fungicidal Concentration of the products tested alone and in combination, it was found that in the case of Candida strains, growth inhibition by limonene occurred at a concentration of 1024 µg/mL. For Fluconazole, growth impairment ranged from > 1024 µg/mL to 256 µg/mL for the strains. And when combined, limonene potentiated the action of FCZ, making fungal colonization unfeasible at concentrations below 1024 µg/mL. Regarding the morphological transition from yeast to hyphae, limonene was used at concentrations of 1024 µg/mL and 512 µg/mL, and it was found that, for CA and CK, the filaments were reduced in number and size at the highest concentration and against CT, the morphological transition from yeast to hyphae/pseudohyphae was totally inhibited, and if compared to the growth control, limonene was able to reduce fungal growth at concentrations greater than 512 µg/mL. This compound has antimicrobial activity described, due to its ability to interfere in the gene expression of the fungus, the limited therapeutic options and the recent emergence of multidrug-resistant Candida species represent a significant challenge for human medicine and highlight the need for new therapeutic approaches, and in this study a great potential of limonene was revealed in relation to the perspective of increasing the efficiency of commercial drug. This work can bring an important contribution to the scientific database, while emphasizing that in-depth studies and tests on the subject, in order to better investigate its effectiveness and mechanisms by which they exert their effects, are still necessary.

RESUMEN

OBJECTIVE: this study evaluated dentin microtensile bond strength (µTBS) and failure modes (at 24 h and one year), bonding interface regarding hybridization, surface morphology regarding demineralization, in situ metalloproteinase (MMP) activity, and antibacterial effect of three dentin etchants compared to 35% phosphoric acid (PA). MATERIALS AND METHODS: The Adper Single Bond 2 adhesive (3 M Oral Care) was applied on moist dentin etched with PA (control) or on air-dried dentin etched with 3% aluminum nitrate + 2% oxalic acid (AN), 6.8% ferric oxalate + 10% citric acid (FO), or 10% citric acid (CA). The µTBS test used 40 human teeth (n = 10). Failure modes and surface morphology were analyzed by scanning electron microscopy (n = 3), while bonding interface morphology and MMP activity were evaluated by laser scanning confocal microscopy (n = 3). Antibacterial activity was evaluated against S. Mutans biofilm by means of viable cells count (CFU/mL). RESULTS: PA presented the highest bond strengths regardless of aging time. PA, AN, and CA showed stable bond strengths after one year of storage. Adhesive and mixed failures were predominant in all groups. Thin hybrid layers with short resin tags were observed for the experimental etchants. The AN-based etchant was able to inhibit MMP activity. All tested etchants presented antibacterial activity against S. Mutans biofilm. SIGNIFICANCE: This study suggests different dentin etchants capable of inhibiting MMP activity while also acting as cavity disinfectants.

RESUMEN

BACKGROUND: Resistance training (RT) consists of planned exercise programs to increase muscle strength capacity through neural and structural adaptations, such as changes in the geometric arrangement of muscle fibers. This study aimed to analyze the influence of RT on muscle architecture in older people. METHODS: This PROSPERO-registered systematic review and meta-analysis (identification number CRD42022340477) followed the PRISMA guidelines. Four electronic databases were searched for eligible randomized controlled trials (RCTs) that observed older individuals submitted to RT programs that reported muscle architecture outcomes. RESULTS: Seventeen RCTs met the eligibility criteria with a total of 488 participants. The main results of the meta-analysis showed that RT interventions had a significant effect on the thickness of the medial gastrocnemius (SMD = 0.12; 95% CI: - 0.07 to 0.17; p < 0.00001; I2 = 0%). CONCLUSION: Based on available evidence, studies included in this review showed improvement in maximum isometric force, pennation angle, fascicle length, thickness, and muscle activation after RT interventions. In turn, the meta-analysis suggested a potential for improving the thickness of the medial gastrocnemius after the intervention. However, any clinical implications drawn from the analyses should be interpreted with caution, as these findings are substantially limited due to a low number of included studies and a potential heterogeneity between studies.

RESUMEN

Background: Acute exercise contributes to decreased feeding through leptin and interleukin/Janus kinase 2/signal transducers and activators of transcription 3 (IL-6/JAK2/STAT3) signaling. Considering the pleiotropic use of substrates by JAK2 and that JAK2 can phosphorylate the Tubby protein (TUB) in CHO-IR cells, we speculated that acute exercise can activate the IL-6/JAK2/TUB pathway to decrease food intake. Aims: We investigated whether acute exercise induced tyrosine phosphorylation and the association of TUB and JAK2 in the hypothalamus and if IL-6 is involved in this response, whether acute exercise increases the IL-6/TUB axis to regulate feeding, and if leptin has an additive effect over this mechanism. Methods: We applied a combination of genetic, pharmacological, and molecular approaches. Key findings: The in vivo experiments showed that acute exercise increased the tyrosine phosphorylation and association of JAK2/TUB in the hypothalamus, which reduced feeding. This response was dependent on IL-6. Leptin had no additive effect on this mechanism. Significance: The results of this study suggest a novel hypothalamic pathway by which IL-6 released by exercise regulates feeding and reinforces the beneficial effects of exercise.

RESUMEN

Aim: To analyze the associations between pain duration, pain levels, anthropometric measures, perceived stress and biochemical markers in women with low back pain. Materials & methods: Forty-two participants were submitted to body mass, height, abdominal circumference, cortisol and creatine kinase (CK) collections. Pain duration, pain levels and stress were analyzed through specific questionnaires. Results: There were positive correlations between abdominal circumference and body mass, duration of pain and age, abdominal circumference, CK and age, CK and BMI, CK and abdominal circumference, and CK and duration of pain (p < 0.05). Conclusion: The higher the level of tissue damage over the years, the greater the tendency for higher levels of low back pain perception. Central fat was related to greater load on the spine.

Lay abstract Low back pain (LBP) is one of the factors that hinders the routine and productivity of women. LBP can cause functional disability and negatively interfere with work activities. The increase in pain can be caused by mechanical postural factors (such as behavioral habits), traumatic (such as accidents), and psychosocial factors (such as physical inactivity). Chronic LBP can influence behavioral aspects due to adaptations of pain feedback, such as postures to avoid pain and sedentary lifestyle. In this study, we observed that duration of LBP was related to higher levels of creatine kinase, which is an enzyme of our body that can be related to tissue damage. Moreover, the abdominal circumference, due to the accumulation of fat in this region, presented correlation with higher levels of tissue damage by creatine kinase and longer exposure time with LBP.

RESUMEN

Aim: To verify the effects of physical exercise on low back pain (LBP) and serum cortisol levels in individuals with chronic LBP. Materials & methods: Randomized controlled trials evaluating the effects of exercise on LBP perception and cortisol levels in adults with nonspecific chronic LBP were included. Results: Four randomized controlled trials were included, with a total of 85 participants in the exercise group and 84 in the control group. The interventions reduced -1.61 (95% CI: -2.36 to -0.85) with inconsistency I2 = 72% (p = 0.031) the LBP level and increased 1.05 (95% CI: 0.22-2.32) with inconsistency I2 = 86% (p < 0.0001) the cortisol levels. Conclusion: The practice of physical exercise for 6 weeks or more reduced LBP levels, whereas the rate of progression of an exercise-training program in people with chronic LBP is greater than 4 weeks, but increased the cortisol serum levels in individuals with LBP.

RESUMEN

PURPOSE: To evaluate the effect of silver diamine fluoride (SDF) application on the microshear bond strength (MBS) of glass-ionomer cements (GIC) to caries-affected dentin (Part 1) and dentin cleaning methods to reduce SDF's potential effect on MBS (Part 2). METHODS: For Part 1, 56 extracted human teeth were randomly divided into eight groups with GIC, 38% SDF application and dentin substrate. Samples of artificial caries-affected human dentin were treated or not with 38% SDF and restored with conventional or resin-modified GIC. The same procedures were performed in sound dentin tested for MBS test after 24 hours. In Part 2, different dentin cleaning agents (water, aluminum oxide, and pumice slurry) were tested after SDF application. The procedure was performed on the group that presented the worst values for MBS in Part 1. Fracture mode was evaluated under scanning electron microscope. Data were statistically analyzed by ANOVA. RESULTS: MBS was affected by the presence of caries and the type of material, with the conventional GIC the most affected (P< 0.05). Pumice slurry was superior in comparison to the other agents in cleaning SDF-treated dentin. Fracture evaluation showed more mixed failures in all the groups. CLINICAL SIGNIFICANCE: Clinicians should have caution when selecting the glass-ionomer cement (GIC) for restorations in silver diamine fluoride (SDF)-treated dentin. The mechanical properties of conventional GIC restorations were more affected than resin-modified GICs. Pumice slurry was the most effective cleaning method to minimize the negative effect of SDF on dentin.

RESUMEN

BACKGROUND: Pharmaceutical nanotechnology represents an efficient alternative for the delivery of pharmacologically active plant-derived compounds, considering their protective capacity, oral bioavailability and drug vectorization capacity. In this context, butters obtained from plant seeds have emerged as promising products for the development of pharmacologically active nanostructures. They possess a complex lipid composition, allowing the formation of different emulsion systems with solid cores, since this mixture of different triglycerides is solid at room temperature and body temperature. Therefore, the systematic mapping around the technological development of nanostructures produced from plant-derived butters is potentially valuable for researchers interested in novel alternative formulations for pharmacological therapy, with potential industrial, economic, health and societal impacts. METHODS: Systematic review was carried out by the search of scientific papers and patents deposited in official databases concerning the development of nanostructured pharmaceutical products using plantderived butters as starting material. The publications obtained were subjected to sorting and analysis by applying the following inclusion/exclusion criteria. RESULTS: The Solid Lipid Nanoparticle (SLN) was the type of nanostructure produced in all the analyzed scientific papers, due to the physicochemical characteristics of the lipid constituents of plantderived butters. In this sense, 54% of the articles have reported the use of Cocoa Butter for the production of nanostructures; 28% for Shea Butter; 6% for Cupuacu Butter, 6% for Murumuru Butter and 6% for Bacuri Butter. DISCUSSION: In the technological prospection, only two patents exhibited SLN as an invention based on cocoa butter and on shea butter, respectively. The production methods employed have included: phase inversion temperature, microemulsion, hot high pressure homogenization, high shear homogenization and ultrasonication. CONCLUSION: In light of this prospective review, the encouragement of novel studies in lipids-based nanotechnology is evident, considering the small number of findings so far, in order to stimulate new research involving plant-derived butters from easily cultivated fruits in tropical regions, then stimulating the pharmaceutical development of new therapeutic alternatives using biocompatible and sustainable raw materials.

RESUMEN

PURPOSE: To assess the effect of silver diamine fluoride (SDF) application on bond strength of current adhesive systems to caries-affected dentin and cleaning procedures to overcome SDF's influence on adhesion to dentin. METHODS: 64 human third molars were randomly divided in eight groups for microshear bond strength testing (MBS). Samples of sound and artificial caries-affected human dentin were treated or not with 38% SDF and restored with an etch-and-rinse or a self-etch bonding system. For the cleaning part, water, aluminum oxide, and pumice paste were used after SDF application to determine whether SDF affected the bond strength to dentin. Fracture mode was evaluated under scanning electron microscope. Data were statistically analyzed by ANOVA. RESULTS: SDF application resulted in the lowest MBS for the self-etching adhesive system on caries-affected dentin (P< 0.05). Cleaning with pumice slurry maintained the MBS in SDF groups when compared to the control groups (not treated with SDF). Fracture evaluation showed more adhesive failures on adhesive systems groups. EDX analysis showed no silver particles when pumice paste was used for cleaning. CLINICAL SIGNIFICANCE: Self-etch adhesive systems do not seem appropriate for bonding SDF-treated dentin restorations. Cleaning SDF-treated dentin with pumice paste reduced the negative effect of SDF on resin-dentin bond strength. Etch-and-rinse adhesive systems seemed not be affected by 38% SDF.

RESUMEN

Pemphigus foliaceus is an autoimmune disease that is sporadic around the world but endemic in Brazil, where it is known as fogo selvagem (FS). Characterized by autoantibodies against the desmosomal cadherin desmoglein 1, FS causes painful erosions, and crusts that may be widespread. The recognition of antigens, including exposed sugar moieties, activates the complement system. Complement receptor 1 (CR1, CD35), which is responsible for the Knops blood group on erythrocytes (York and McCoy antigens), is also expressed by antigen-presenting cells. This regulates the complement system by removing opsonized antigens, blocking the final steps of the complement cascade. Membrane-bound CR1 also fosters antigen presentation to B cells, whereas soluble CR1 has anti-inflammatory properties. CR1 gene polymorphisms have been associated with susceptibility to complex diseases. In order to investigate the association of CR1 polymorphisms with FS susceptibility, we developed a multiplex sequence-specific assay to haplotype eleven polymorphisms in up to 367 FS patients and 242 controls from an endemic area and 289 from a non-endemic area. We also measured soluble CR1 (sCR1) in the serum of 53 FS patients and 27 controls and mRNA levels in the peripheral blood mononuclear cells of 63 genotyped controls. The haplotypes CR1*3B2B (with the York antigen-encoded by p.1408Met) and CR1*3A2A (with p.1208Arg) were associated with protection against FS (OR = 0.57, P = 0.027, and OR = 0.46, P = 0.014, respectively). In contrast, the CR1*1 haplotype (with the McCoy antigen - encoded by p.1590Glu) was associated with FS susceptibility (OR = 4.97, P < 0.001). Heterozygote rs12034383*A/G individuals presented higher mRNA expression than homozygotes with the G allele (P = 0.04). The lowest sCR1 levels occurred in patients with active disease before treatment (P = 0.036). Patients in remission had higher levels of sCR1 than did healthy controls (P = 0.013). Among those under treatment, patients with localized lesions also presented higher sCR1 levels than those with generalized lesions (P = 0.0073). In conclusion, the Knops blood group seems to modulate susceptibility to the disease. Furthermore, corticosteroid treatment might increase sCR1 serum levels, and higher levels may play an anti-inflammatory role in patients with FS, limiting the distribution of lesions. Based on these results, we suggest CR1 as a potential new therapeutic target for the treatment of FS.

RESUMEN

Thiourethane oligomers added to methacrylate matrices improve fracture toughness and reduce polymerization stress. In this study, the oligomers are added to the surface of inorganic fillers in the formulation of resin composites. Systematically varied fillers treated either with the thiourethane or a methacrylate silane control are tested. Thiourethane reduces the rate of polymerization of light-cured composites but does not affect the final degree of conversion (DC). Filler functionalization with thiourethane increases the depth of polymerization, in filler type-dependent fashion. Thiourethane reduces the polymerization stress for all fillers. The findings suggest that this approach results in the same general effects with the addition of thiourethanes directly to the matrix. This is accomplished with a lower overall concentration of thiourethane, and with no prejudice to the handling characteristics of the material.

RESUMEN

OBJECTIVES: This study evaluated the efficacy of a thiourethane(TU)-modified silane agent in improving properties in filled composites. METHODS: The TU-silane agent was synthesized by combining 1,3-bis(1-isocyanato-1-methylethyl)benzene and 3-(triethoxysilyl)propyl isocyanate with trimethylol-tris-3-mercaptopropionate (TMP), at 1:2 isocyanate:thiol, leaving pendant thiol and alkoxy silane groups. Barium glass fillers (1µm average particle size) were functionalized with 5wt% TU-silane in an acidic ethanol solution. Commercially available 3-(trimethoxysilyl)propyl methacrylate (MA-silane) and (3-mercaptopropyl)trimethoxysilane (SH-silane), as well as no silane treatment (NO-silane), were used as controls. Composites were made with BisGMA-UDMA-TEGDMA (5:3:2), camphorquinone/ethyl-4-dimethylaminobenzoate (0.2/0.8wt%) and di-tert-butyl hydroxytoluene (0.3wt%) and 70wt% silanated inorganic fillers. Polymerization stress (PS) was measured using a cantilever beam apparatus (Bioman). Methacrylate conversion (DC) and rate of polymerization (RP) during photoactivation (800mW/cm2) were followed in real-time with near-IR. Flexural strength/modulus (FS/FM) were evaluated in three-point bending with 2×2×25 mm. STATISTICAL ANALYSIS: 2-way ANOVA/Tukey's test (α=5%). RESULTS: DC, Rpmax and E were similar for all groups tested. FS was similar for the TU- and MA-silane, which were statistically higher than the untreated and SH-silane groups. Stress reductions in relation to the MA-silane were observed for all groups, but statistically more markedly for the TU-silane material. This is likely due to stress relaxation and/or toughening provided at the filler interface by the oligomeric TU structure. SIGNIFICANCE: TU-silane oligomers favorably modified conventional dimethacrylate networks with minimal disruption to existing curing chemistry, in filled composites. For the same conversion values, stress reductions of up to 50% were observed, without compromise to mechanical properties or handling characteristics.

RESUMEN

The aim of this study was to synthesize and characterize mesoporous materials SBA-15 and SBA-15 modified with 3-(methacryloxy)-propyl-trimethoxysilane (MPS) to be used as inorganic filler in restorative dental composites and adhesives, and evaluate the main physical-chemical properties of the resulting material. The SBA-15 and SBA-15/MPS were characterized by FTIR, BET and X-Ray and combined with TEGDMA, bis-GMA and commercial spherical silica to produce dental composites. Afterwards, the mesoporous materials were combined with TEGDMA, bis-GMA and HEMA to make adhesives. To compare the results, composites and adhesives containing only commercial spherical silica were investigated. Some physical-chemical properties such as degree of conversion (DC), flexural strength (FS) and modulus (FM), water sorption and solubility (Wsp and Wsl), specific area (BET), and the leachable components were evaluated. The SBA-15/MPS can be used to prepare dental restorative materials, with some foreseeable advantages compared with pure SBA-15 dental materials and with improved properties compared with commercial spherical silica dental materials. An important improvement was that the dental materials based on modified SBA-15 presented a reduction of approximately 60% in leaching of unreacted monomers extracted by solvent compared to the control group.

RESUMEN

The aim of this in vitro study was to synthesize three new methacrylate monomers based on the modification of saccharides structures (glucose-Gluc, sucrose-Sucr and chitosan-Chit) with glycidyl methacrylate, and to use them in the composition of dental adhesives. Three methacrylate saccharide monomers were synthesized and characterized by mid-IR, 1H and 13C NMR, antioxidant activity and cytotoxic effect. Monomers included: one monosaccharide - Gluc-MA; one disaccharide - Sucr-MA; and one polysaccharide - Chit-MA. Primers containing HEMA, methacrylate saccharide monomers at concentrations of 0 (control), 1, 2 or 4 wt%, 60 wt% ethanol aqueous solution (pH3.0) and initiator system were formulated. Primers were used in conjunction with a bond step and composite paste to restore caries-free third molars, and dentin bond strength (24 hours and 6 month of storage in water), and antimicrobial activity (Alamar Blue test) were tested. Degree of conversion (DC) and maximum rate of polymerization (Rpmax) of the primers themselves were also analyzed. The mid-IR, 1H and 13C spectrum confirmed the presence of vinyl group on the structure of saccharides. Chit-MA showed low antioxidant activity and did not present a cytotoxic effect. Gluc-MA and Sucr-MA possess antioxidant and cytotoxic activity, concentration dependent. In the presence of methacrylate saccharide monomers, the primers showed DC comparable to the control group, except Gluc-MA4%, Sucr-MA4% and Chit-MA1%, which showed a range of 64.6 from 58.5 %DC. Rpmax was not statistically different for all the groups (p = 0.01). The bond strength of Sucr-MA1% increased from 25.7 (±2.8) to 40.6 (±5.3) MPa after 6 months of storage. All the synthesized monomers showed some antimicrobial activity after polymerization. Gluc-MA and Chit-MA 4% and Sucr-MA 1, 2 and 4% led to decrease bacterial metabolism. Sucr-MA 1% showed better results regarding the decrease in bacterial metabolism and increasing the bond strength after 6 months of storage.

RESUMEN

OBJECTIVES: This study evaluated three different sterilization/disinfection techniques for resin composites on bacterial growth and surface modification after decontamination. METHODS: Two resin composites were sterilized/disinfected with three different techniques: UV light, 1% chloramine T, and 70% ethanol. Four different times were used for each technique to determine the shortest time that the solution or UV light was effective. The influence of sterilization/disinfection technique on bacterial growth was evaluated by analyzing the metabolic activity, using the AlamarBlue™ assay, bacterial viability, and SEM images from biofilms of Streptococcus mutans. The surface change, after the process, was analyzed with ATR/FTIR and SEM images. The solutions used for decontamination (1% chloramine-T and 70% ethanol) were analyzed with 1 H-NMR to identify any resin compounds leached during the process. RESULTS: One minute of decontamination was efficient for all three methods tested. Chloramine-T increased the surface porosity on resin composites, no changes were observed for UV light and 70% ethanol, however, 1 H-NMR identified leached monomers only when 70% ethanol was used. No chemical change of the materials was found under ATR/FTIR analyses after the decontamination process. Chloramine-T, with no previous wash, increased the bacterial viability for both resin composites and increased the bacterial metabolism for the resin composite without fluoride. CONCLUSION: UV light had no interference on the resin composites properties tested using 1 min of exposure compared to the other decontamination methods. © 2017 Wiley Periodicals, Inc. RESUMEN

OBJECTIVE: To determine the prevalence of the types of relations to career - commitment and entrenchment - of nurses from public and private hospitals. METHODS: Field survey with a quantitative approach. A total of 237 nurses participated in the survey through a questionnaire with 43 questions about sociodemographic data, occupation, and relation to career. Descriptive statistics, frequency analysis, and Mann-Whitney U test were used for data analysis. RESULTS: Nurses from public and private hospitals have a high level of career commitment and a median level of career entrenchment. CONCLUSIONS: Nurses have a stronger relation of affection and identification to career than a relation of stagnation, maintained primarily due to investments, followed by emotional costs and lack of alternatives. This fact is associated with individuals continuing in nursing and having a strong commitment to activities.

RESUMEN

The aim of this study was to estimate the prevalence and factors associated with the occurrence of incidents related to medication, registered in the medical records of patients admitted to a Surgical Clinic, in 2010. This is a cross-sectional study, conducted at a university hospital, with a sample of 735 hospitalizations. Was performed the categorization of types of incidents, multivariate analysis of regression logistic and calculated the prevalence. The prevalence of drug-related incidents was estimated at 48.0% and were identified, as factors related to the occurrence of these incidents: length of hospitalization more than four days, prescribed three or more medications per day and realization of surgery intervention. It is expected to have contributed for the professionals and area managers can identify risky situations and rethink their actions.
